metadata.dc.description.resumo: O seguinte trabalho tem como intuito descrever o Estágio Curricular Supervisionado, o qual fora orientado pelo professor Dr. José Carlos Ribeiro Júnior e supervisionado pela professora Dra. Alice Fernandes Alfieri. Todo o estágio foi feito no Laboratório de Virologia Animal na Universidade Estadual de Londrina entre 15 de agosto e 27 de outubro de 2022. Além de apresentar o local, seus componentes, equipe, exames oferecidos e relatar as atividades desenvolvidas no decorrer do estágio, esta monografia tem como propósito abordar o diagnóstico molecular de enfermidades causadas por vírus que originam diarreia em bovinos. Através de técnicas que utilizam da biologia molecular para melhor acurácia em identificar patógenos, material genético fora extraído das amostras que em seguida foram levadas para análise com eletroforese em gel de poliacrilamida e RT-PCR seguida de eletroforese em gel de agarose e leitura em luz ultravioleta. Entre as amostras enviadas para análise no laboratório, houve uma grande quantidade de fezes diarreicas, portanto serão descritas as técnicas, resultados e discussões feitas a partir delas.
Abstract: The following work aims to describe the supervised curricular internship, which was guided by professor Dr. José Carlos Ribeiro Júnior and supervised by professor Dr. Alice Fernandes Alfieri. The entire internship was completed in the Animal Virology Laboratory at the State University of Londrina, between the periods of August 15 and September 27, 2022. Besides introducing the premises, its components, the team, exams offered and report the activities developed during the internship, this essay also aims to address the molecular diagnosis of infirmities caused by viruses that originates diarrhea in bovines. Through procedures in which uses molecular biology to improve the accuracy to identify pathogens, genetic material was extracted from samples that later were analyzed by polyacrylamide gel electrophoreses and underwent RT-PCR followed by electrophoreses in agaroses gel and reading through UV light. Among the samples that were sent to the lab was a large amount of diarrheal feces, therefore it will be describe the techniques, the results and the discussions made from them.
