metadata.dc.description.resumo: O mosquito Aedes aegypti L. (Diftera: Culicidae) é o principal vetor do vírus da dengue (DENV), febre amarela, Zika e Chikungunya causando várias epidemias em países tropicais e subtropicais. Neste contexto, o objetivo deste trabalho foi avaliar a composição química dos extratos de Tabebuia heptaphylla, Chiococca alba e Peltastes peltatus, e a atividade entomocida, de repelência e antidegue. Analisamos a toxicidade de diferentes extratos de T. avellanedae obtidos por Soxhlet e Maceração contra larvas de 3º instar de A. aegypti. Nos ensaios de toxicidade dos extratos de T. heptaphylla observa-se que os extratos acetona e acetato de etila obtidos por Soxhlet foram mais tóxicos contra larvas de 3º instar de A. aegypti, with CL50 of 100.1 and 151.0 μg/mL, respectivamente. Neste contexto, em todos os casos, os extratos de T. heptaphylla apresentaram 100% de mortalidade contra larvas de 3º instar após um período de 12 horas. Enquanto nos ensaios de repelência de T. heptaphylla, cinco tipos de concentrações (0,2; 0,4; 2.0; 4.0; 8.0 mg/cm2) foram testadas e as duas formulações repelentes (Gel e Creme) incorporadas ao extrato metanólico na concentração de 4,0 mg/cm2 apresentaram maior proteção (100%) durante 3 horas de exposição. Para teste de repelência com o extrato metanólico de C. alba, a formulação com propilenoglicol e álcool a 70%, apresentou 100% de proteção durante 3 horas. Além disso, vários compostos químicos identificados nos extratos clorofórmico e hexânico de T. heptaphylla, como naphthoquinone2_6_di_tert_butylnaphthalene (lapachol) e 2_2_dimethyl_2H_naphtho[2_3_b]pyran_5_10_dione apresentaram interação com o receptor da proteína de ligação a odores do A. aegypti (AaegOBP1). Assim também foi envidenciada a interação desse receptor com os compostos químicos rutina, álcool oleílico, trifluoroacetato, ácido 3,5-dimetoxicinâmico e ácido (E) -4-metoxicinâmico presentes no extrato de C. alba, Em relação a atividade anti-dengue de C. alba foram realizados testes contra vírus DENV-2. O extrato hexânico com 6.25 μg/μL mostrou redução no título viral de ≅ 39%. Enquanto o extrato acetona de Peltastes peltatus com 50 μg/μL apresentou uma redução no título viral de ≅ 55,63%. Os resultados foram superiores ao composto sintético antidengue, ribavirina, nas concentrações 146.5ug/uL que apresentou redução de títulos virais de 49,19%. Baseado nestes resultados, os extratos de T. heptaphylla e C. alba podem ser utilizados no desenvolvimento de repelentes para A. aegypti. E ao mesmo tempo esses extratos e o extrato de Peltastes peltatus podem ser potencialmente usados para prospectar formulações contra o vírus DENV- 2.
Abstract: The mosquito Aedes aegypti L. (Diftera: Culicidae) is the primary vector of dengue, yellow fever, Zika virus and Chikungunya, responsible for several epidemics in tropical and subtropical countries. Therefore, the objective of this study was to evaluate the chemical composition of the extracts of T. heptaphylla, Chiococca alba and Peltastes peltatus, as well as their entomocidal activities, repellency and anti-dengue activity. We analyzed the toxicity of T. avellanedae extracts obtained by Soxhlet and maceration against 3rd instar larvae of A. aegypti. In toxicity assays of T. avellanedae extracts, we observed that the acetone and ethyl acetate extracts obtained by Soxhlet were more toxic against 3rd instar larvae of A. aegypti, with LC50 of 100.1 and 151.0 μg/mL, respectively. In this context, in all cases, T. avellanedae extracts showed 100% mortality with 3rd instar larvae after 12 hours. In the T. heptaphylla repellency assays, five concentrations (0.2, 0.4, 2.0, 4.0, 8.0 mg/cm2) were tested and two repellent formulations were prepared (gel and cream) to be incorporated into the extract methanol concentration at 4.0 mg/cm2 presented the highest protection (100%) for a period of 3 hours. For repellency test with the methanolic extract of C. alba, the formulation with propylene glycol and alcohol at 70% provided 100% protection for 3 hours. Furthermore, various chemical compounds identified in the chloroform and hexanic extracts of T. heptaphylla, including naphthoquinone 2_6_di_tert_butylnaphthalene (lapachol) and 2,2_dimethyl-2H-naphth [pyrazine] pyran-5_10 interacted with the A. aegypti (AaegOBP1) odor-binding protein receptor. Thus, the interaction of this receptor with the chemical compounds rutin, oleyl alcohol, trifluoroacetate 3,5-dimethoxycinnamic acid, and (E)-4- methoxycinnamic acid present in the extract of C. alba was evidenced to anti-dengue activity the reduction in viral titer of ≅ 39% was observed against DENV-2, while the acetone extract of Peltastes peltatus at 50 μg/μL showed a reduction in viral titer of ≅ 55.63% The results were greaterthan for the synthetic anti-dengue compound, ribavirin, at 146.5 μg/μL concentrations, and showed a reduction of 49.19% in viral titers. Based on these results, extracts of T. heptaphylla and C. alba can be used in the development of A. aegypti repellents, and at the same time these extracts and the Peltastes peltatus extracts may potentially be used to prospect formulations against the DENV-2 virus.
